The experimental procedure referred to the LanthaScreen Eu Kinase Binding Assay Screening Protocol and Assay Conditions provided by ThermoFisher Scientific.B-1. Material Preparation:3-fold serial dilution was performed on the test compound stock solution (1 mM) prepared in 100% DMSO was for 10 times for ready to use to determine the IC50 of the test compound on MYLK-4. Kinase/Antibody Mixture was pre-diluted with Kinase Buffer to 2× working concentration. B-2. Analytical Method 1. 3.84 μL of kinase buffer was added to a white 384-well flat plate (Greiner, Cat. NO. 784207), then 8.0 μL of the kinase/antibody mixture mentioned above was added thereto, and then 4.0 μL of the tracer (AlexaFluor labeled Tracer) was added thereto, and after that 160 nL of the above diluted test compound at an appropriate concentration was added thereto, and the culture plate was shaken for 30 seconds. 2. Next, the culture plate was placed at room temperature and incubated for 60 minutes, and then the data was read and analyzed with a fluorescence plate reader.
